Shoot Proliferation, Rooting, and Genetic Stability of Six Micropropagated Olive (Olea europaea L.) Genotypes

This research aimed to explore the effect of zeatin (6-γ,γ-dimethylallyl-amino) concentration on in vitro shoot proliferation six olive (Olea europaea L.) cultivars, ‘Aggizi Shami,’ ‘Coratina,’ ‘Frantoio,’ ‘Manzanillo,’ ‘Picual,’ and ‘Toffahi.’ The cultivars were reproduced via multiplication Rugini medium (OM) with three concentrations (2, 4, 6 mg L-1) for subcultures. Shami’ ‘Toffahi’ had highest numbers. Increasing improved propagation. best response was recorded OM supplemented at mg·L-1; this treatment resulted high values numbers shoots, leaves, internodes. Then, rooting ability shoots analyzed using two different methods. regenerated successfully rooted ½OM indole-3-butyric acid (IBA), ‘Coratina’ ‘Picual’ having percentages. methods genotype-dependent. Rooting IBA more successful than dipping IBA. Based growth potential most adaptable conditions, followed by ‘Frantoio’ ‘Manzanillo.’ recalcitrant as they exhibited low potential. After subcultures, genetic stability determined. “Manzanillo” showed index (96%), while lowest value (85%), based start codon targeted (SCoT) data analysis.
